Die Pigeon Circovirus (PiCV) DNA-Test Kit is a specialized qPCR-based molecular diagnostic tool designed to detect PiCV, a major cause of young pigeon disease syndrome (YPDS) and immune suppression in racing and breeding pigeons. PiCV replicates rapidly in immune system cells, leading to weakened immunity, low lymphocyte counts, and increased susceptibility to secondary infections.

Kit Components:

PCR Reaction Setup:

• Reaction volume: 25 µL

• Mix: 23 µL Detection Reagent + 2 µL Sample DNA / Positive / Negative Control

• Real-time PCR cycling conditions:

  • 50°C for 2 min (UNG digestion)

  • 95°C for 5 min (denaturation)

  • 40 cycles of:

    • 95°C for 10 sec

    • 60°C for 35 sec (fluorescence read in FAM channel)

Compatible with ABI series and other qPCR instruments. ROX reference dye optional if needed.

Interpretation der Ergebnisse:

• Positiv: Ct ≤ 33 with typical “S” curve

• Verdächtiger: 33 < Ct < 38 (retest recommended to rule out inhibitors or contamination)

• Negativ: Ct ≥ 38 or no amplification curve

Sample Requirements:

• Abstriche: Oral or cloacal swabs placed into sterile centrifuge tubes

• Blut: EDTA anticoagulated whole blood (avoid heparin)

• Tissue: Fresh liver tissue (≤100mg), homogenized in sterile water

DNA extraction should be performed with a validated kit such as DP202. Extracted DNA should be stored at 4°C (≤7 days) or –20°C (≤6 months).
