Kit Components:

PCR Reaction Setup:

• Reaction volume: 25 µL

• Mix: 23 µL Detection Reagent + 2 µL Sample DNA / Positive / Negative Control

• Real-time PCR cycling conditions:

  • 50°C for 2 min (UNG digestion)

  • 95°C for 5 min (denaturation)

  • 40 cycles of:

    • 95°C for 10 sec

    • 60°C for 35 sec (fluorescence read in FAM channel)

Compatible with ABI series and other qPCR instruments. ROX reference dye optional if needed.

Result Interpretation:

• Positive: Ct ≤ 33 with typical “S” curve

• Suspect: 33 < Ct < 38 (retest recommended to rule out inhibitors or contamination)

• Negative: Ct ≥ 38 or no amplification curve

Control results must validate test effectiveness. Follow the manual strictly.

Sample Requirements:

• Swabs: Oral or cloacal swabs placed into sterile centrifuge tubes

• Blood: EDTA anticoagulated whole blood (avoid heparin)

• Tissue: Fresh liver tissue (≤100mg), homogenized in sterile water

DNA extraction should be performed with a validated kit such as DP202. Extracted DNA should be stored at 4°C (≤7 days) or –20°C (≤6 months).
