PCR Test Workflow

1. Sample Preparation

Use a DNA extraction kit such as SanshiBio DP202 to extract genomic DNA from bird feathers, blood, or tissues.

2. Reaction Setup

• Mix: 7.5 µL Reagent C + 10 µL Reagent D + 1 µL Reagent E per reaction.

• Add 2 µL DNA sample.

• Total reaction volume: 20 µL.

3. PCR Program

• Initial Denaturation: 94°C, 3 min

• 32 cycles:

  • Denature: 94°C, 30 sec

  • Anneal: 50°C, 45 sec

  • Extend: 72°C, 50 sec

• Final Extension: 72°C, 8 min

• Hold: 4°C

4. Gel Electrophoresis

Use 1%–1.5% agarose gel in TAE or TBE buffer to visualize the PCR bands:

• Single ~700 bp band: Male

• ~700 bp and ~400 bp or only 400 bp: Female

• No band: Retest or consult technical support

Important Notes

• Keep separate areas for sample prep, reaction setup, and product analysis to avoid contamination.

• Wear gloves and lab coats; sterilize tools regularly.

• Do not open PCR tubes immediately after amplification to prevent aerosol contamination.
