Utilizing a non-column, enzyme-based lysis protocol, the kit eliminates the need for centrifugation columns or hazardous solvents, enabling rapid sample preparation in as little as 20 minutes. Protocol Summary

⚠️ Equipment Required: PCR machine or metal block heater (55°C & 95°C)

1. Prepare Digestion Buffer: Mix 50 µL Reagent A + 2.5 µL Reagent B per sample. Dispense 52 µL into PCR tubes.

2. Add Sample:

   • フェザー: Cut finely at the shaft; place into the buffer.

   • Blood/Saliva: Add ≤10 µL directly to buffer.

   • Mouse Tissue: Mince ≤0.5 cm piece and mix well.

3. Incubate:

   • 55°C for 10 min (lysis step)

   • 95°C for 5 min (enzyme inactivation)

4. Cool & Centrifuge:

   • Chill at -20°C for 5 min or at 4°C for 15 min.

   • Briefly centrifuge and use the supernatant for PCR.

💡 Extracted DNA can be stored at 4°C for 1 month or -20°C for up to 1 year.

• Mix all reagents thoroughly before use.

• Do not open PCR tubes immediately after high-temperature incubation.
