Product Overview

This molecular diagnostic kit is specially developed for the detection of Psittacine Beak and Feather Disease Virus (PBFDV) in a wide range of avian sample types, including feathers, blood, tissues, oral swabs, and cloacal swabs. The kit utilizes real-time fluorescence PCR (qPCR) technology to amplify and detect the highly conserved gene region of PBFDV, ensuring high specificity and sensitivity.

Incorporated with an endogenous internal reference gene, the kit monitors both the nucleic acid extraction process and PCR amplification quality, effectively minimizing false negatives and improving the overall reliability of results.

Kit Components

Almacenamiento: Store at -20°C; shelf life ≥ 12 months.

Sample Requirements

• Plumas: At least 5 fresh feathers (with shaft) from chest, abdomen, or leg. Do no use naturally shed feathers.

• Sangre: EDTA-preserved; store at 2–8°C (≤7 days) or -20°C (≤3 months). Avoid heparin and repeated freeze-thaw cycles.

• Tejido: ≤100mg soft organ or muscle tissue (no skin/fascia); homogenize in sterile water.

• DNA Extraction: Use a validated kit such as Three Lions Biotech Animal Genomic DNA Rapid Extraction Kit (DP202).

Workflow Summary

1. Sample Processing

   • Extract nucleic acids per standard kit protocols.

   • Store DNA on ice during preparation and testing.

2. Reaction Setup

   • Thaw all reagents, centrifuge briefly.

   • Add 2μL of sample DNA, positive or negative control, into 23μL reaction mix (final volume: 25μL).

   • Mix and centrifuge before loading onto the PCR machine.

3. PCR Amplification

   • Pre-denaturation: 95°C for 3 min

   • 35 Cycles of:

     • Denaturation: 95°C for 10 sec

     • Annealing/Extension: 60°C for 40 sec

     • Fluorescence detection at 60°C

   • Channels: FAM (PBFDV target), VIC/HEX (internal control)

4. Result Interpretation

   • Valid test: Positive control Ct < 30 with S-curve; negative control Ct ≥ 35 or no curve.

   • Internal control (VIC): Ct ≤ 32 = valid sample.

   • PBFDV detection (FAM):

     • Ct ≤ 32: Positive

     • 32 < Ct < 35: Suspect – retest

     • Ct ≥ 35 or no curve: Negative

Precauciones

• Conduct all steps in designated clean areas to prevent cross-contamination.

• Wear gloves and lab coats; use separate pipettes for pre- and post-PCR.

• Do not open PCR tubes immediately after reaction; allow them to cool.

• Reagents are for research use only. Not intended for clinical diagnosis.
