Product Overview:

The Pigeon Adenovirus (PiADV) DNA Test Kit is a sensitive and specific qPCR diagnostic tool designed for the detection of PiADV, a highly contagious virus that infects pigeons, parrots, and other birds. PiADV is a major cause of acute diarrhea in young pigeons and spreads rapidly through both vertical transmission (via eggs) e horizontal contact.

There is no effective antiviral treatment currently available. Therefore, early and accurate detection using this PiADV nucleic acid detection kit is crucial for timely disease management and for reducing viral spread within bird populations.

This kit utilizes optimized primers and fluorescent probes targeting conserved PiADV gene sequences. It enables precise, real-time qPCR testing from oral/cloacal swabs, whole blood, and tissue samples.

Kit Contents:

Sample Types & Requirements:

• Swabs: Sterile oral or cloacal swabs, stored at 2–8°C (≤24h) or –20°C (if delayed)

• Blood: EDTA-treated whole blood (not heparin), fresh or stored ≤7 days at 2–8°C or ≤3 months at –20°C

• Tissue: ≤100 mg fresh liver homogenized in 200–500 µL sterile water

Nucleic acids should be extracted using DP202 or equivalent extraction kits. Extracted DNA can be stored ≤7 days at 4°C or ≤6 months at –20°C.

qPCR Reaction Setup:

1. Thaw reagents completely and centrifuge briefly.

2. For each test sample + 2 controls (positive and negative), prepare reaction tubes:

   • Add 23 µL PiADV Detection Mix

   • Add 2 µL of DNA sample, positive ou negative control

   • Total volume: 25 µL per tube

PCR Cycling Parameters:

Compatible with ABI and other real-time PCR systems. ROX dye optional if required.

Result Interpretation:

• Positive: Ct ≤ 33 with “S” curve

• Suspect: Ct 33–38 – retest recommended

• Negative: Ct ≥ 38 or no amplification

Controls must meet validity criteria:

• Positive Control Ct < 28

• Negative Control: Ct ≥ 38 or undetected

Important Notes:

1. Strictly separate sample prep, reagent handling, and amplification zones to prevent contamination

2. Wear gloves and lab clothing; change regularly

3. Do not open PCR tubes immediately after run – allow to cool

4. Avoid repeated freeze-thaw of reagents
